COMPARISON OF PREGABALIN AND NORTRIPTYLINE ON SLEEP, QUALITY OF LIFE IN POSTHERPETIC NEURALGIA : A PROSPECTIVE, RANDOMISED, OPEN LABEL, PARALLEL STUDY
Introduction- Postherpetic neuralgia (PHN) is a complication of herpes zoster and severe pain affects sleep and quality of life . Most frequently anticonvulsants ( gabapentin or pregabalin ) and tricyclic antidepressants (amitriptyline or nortriptyline) are used as systemically in management . Aim of study: was to study and compare effect of Pregabalin and Nortriptyline on sleep and quality of life in patients of PHN.
Materials and methods The present study was conducted in 48 patients of PHN attending OPD of Dermatology, GMC Jammu , Based on inclusion/exclusion criteria patients were randomized into two treatment arms Group A (pregabalin 150mg/day )and group B ( amitriptyline 25mg /day) and were followed up to eight weeks and assessed for Sleep (MOS) and quality of life( EQ-5D-5L score) .Clinicians Global Impression of Change (CGIC )score and Patient Global Impression of Change(PGIC) score were used to evaluate overall clinical improvement.
Results: ,Both drugs significantly improved all parameter(p<0.0001) . However, on comparison pregabalin was found to be better than Nortriptyline at eight weeks on mean CGIC((p<0.004), PGIC((p<0.0003), and was numerically better on MOS and Mean EQ-5D-5L .
Conclusion: Both pregabalin and nortriptyline were equally effective in improving sleep and quality of life . Overall clinical improvement was better with pregabalin than nortriptyline at eight weeks .
